2. Set up two bins of water: Room temperature (25C) and 10C (Ice added). Get 3 Respirometers
3. Respirometer 1: Fill graduated cylinder with 50 mL of water. Add 25 GERMINATING PEAS and determine amount of water displaced. Therfore, the volume of the peas is calculated. Remove peas and place on paper towel.
4. Respirometer 2: Fill graduated cylinder with 50 mL of water and add 25 DRIED PEAS/NONGERMINATING into cylinder and add glass beads until this volume is equal to the volume of germinated peas in the previous respirometer. Remove peas and beads and place on paper towel.
5. Respirometer 3: Fill graduated cylinder with 50 mL of water and add enough glass beads that their volume is equal to the volume of the germinating peas in Respirometer 1. Remove peas and place on paper towel. *Repeat steps 1-4 so you can make a second set of peas to be used in Respirometers 4, 5, 6. *PEAS AND/OR BEADS ON THE PAPER TOWELS WILL BE POURED INTO THE RESPIROMETERS RESPECTIVELY.
6. ASSEMBLING RESPIROMETERS. Get 6 vials complete with a stopper and pipette. Put a small piece of cotton at the bottom of each vial and use a dropper to dampen the cotton with 15% of POTASSIUM HYDROXIDE (KOH). The respirometers must be dry on the inside, otherwise the results will be skewed. Place a piece of NONABSORBENT COTTON above the KOH cotton, to prevent the peas from touching the potassium hydroxide.
***TIP: THE AMOUNT OF COTTON WITH THE KOH SHOULD BE CONSTANT (SAME) FOR ALL SIX RESPIROMETERS!!***

7. Add the FIRST SET of peas and/or beads to one of their designated vials. Do the same with the second set, but add them to respirometers 4, 5, 6 respectively. You must insert the stopper with the calibrated pipette to prevent leakage of substances into or out of the vial.
8. Create two "bridges" of masking tape (One long piece each) and attach each end of it to the opposite sides of each of the water bins. Remember to place Vials 1, 2, and 3 in the 25C container, with the ends resting on the tape. Vials 4, 5, and 6 should be sitting on the tape above the 10C container.
9. Let the vials sit in their respective containers for 7 minutes until they reach equilibrium and then immerse the vials into the water. The water around the pipettes will rush into them, then stop after a couple of seconds. THE PIPETTES SHOULD BE ABLE TO BE READ THROUGH THE WATER THROUGHOUT THE EXPERIMENT. Try not to move the pipettes or let the water become contaminated with debris. The water should also maintain a constant temperature throughout the experiment.
10. Wait 3 more minutes to allow the respirometers to equilibrate and record the original water level in each pipette. Also remember to check the temperature of the water to see if it is still constant. Check every 5 minutes for 20 minutes to record the water levels in each pipette in the data table.
