EXPERIMENT
MICROBIOLOGICAL EXAMINATION OF WATER
Objective(s):
 To determine if the water samples are safe and potable.
 To determine if the water samples are contaminated.
 To determine if the institution abides with the standards and requirements of the
Philippine National Standards for Drinking Water.
 To familiarize the students about water testing.
Intended Learning Outcomes (ILOs):
At the end of this study, the students shall be able to:
 Determine the function and the importance of examining the potability of water.
 Determine the sampling of both filtered and unfiltered or in other term (before and
after) water samples.
 Compare the result of both before and after water examination samples.
Discussion:

Water quality refers to the chemical, physical, biological, and radiological characteristics of
water. It is a measure of the condition of water relative to the requirements of one or more
biotic species and or to any human need or purpose. It is most frequently used by reference
to a set of standards against which compliance, generally achieved through treatment of the
water, can be assessed.

Public water systems are required to deliver safe and reliable drinking water to their
customers 24 hours a day, 365 days a year. If the water supply becomes contaminated,
consumers can become seriously ill. Fortunately, public water systems take many steps to
ensure that the public has safe, reliable drinking water. One of the most important steps is
to regularly test the water for coliform bacteria.

Coliform bacteria are organisms that are present in the environment and in the feces of all
warm-blooded animals and humans. Coliform bacteria will not likely cause illness. However,
their presence in drinking water indicates that disease-causing organisms (pathogens) could
be in the water system. Most pathogens that can contaminate water supplies come from the
feces of humans or animals. Testing drinking water for all possible pathogens is complex,
time-consuming, and expensive. It is relatively easy and inexpensive to test for coliform
bacteria. If coliform bacteria are found in a water sample, water system operators work to
find the source of contamination and restore safe drinking water.

Total coliform, fecal coliform, and E. coli are all indicators of drinking water quality. The total
coliform group is a large collection of different kinds of bacteria. Fecal coliforms are types of
total coliform that mostly exist in feces. E. coli is a sub-group of fecal coliform.

Resources/Instruments Required:

 Water (from TIP Manila 5th floor faucet and 7th floor drinking fountain)
 Sterilized Bottles
 Water Testing Institution

Procedure:

1. Accommodate with water testing institutions that are accredited and recognized by
the National Health Department.

2. The water testing institutions will provide sterilized bottles.

3. Choose your sample sources.

4. Before getting water samples, make sure to disinfect your hands.

5. Acquire water samples from the chosen sample sources.

6. Avoid coughing or sneezing near your acquired water samples.

7. Deliver the water samples to the water testing institutions.

Observations:

When we acquired the water samples from both sample sources, the water is clear and looks
safe. We avoid coughing or sneezing near the water samples as required.

Conclusion:

Based on the test results, the water sample from the 5th floor faucet and 7th floor drinking
fountain are both clear of physical characteristics, have less than 1.1 MPN/100 ml total
coliform and MPN/100 ml fecal coliform, and less than 500 cfu/ml heterotrophic plate count
which means that both water samples passed the national standards for drinking water. It
also means that the water from sample sources are potable and safe.

Recommendations:

To be sure about the safety and potability of domestic or public water, we recommend to
give them microbiological examination. It is really helpful for citizens and will help avoid
health accidents.

When acquiring water samples for testing, make sure to follow the required procedure by
the testing institutions for acquiring water.
