PREPARATION OF RED CELL SUSPENSION, DILUTION AND
CONCENTRATION

OBJECTIVES:
At the end of this activity, the student should be able to:

Reagents, Supplies and Equipment:

Procedure:
A. Wearing gloves, collect blood through venipuncture and transfer it in an EDTA tube.
B. Centrifuge, separate the plasma and discard the buffy coat.

C. Preparation of EXACT 5% Washed Red Cell Suspension


1. Using a transfer pipette, obtain some red cells from the bottom of the original tube and
deliver 10 drops into Tube #2 labeled “Washed cells”.
2. Add normal saline ( NSS ) into the tube labeled “Washed Cells” up to 3/4 level of the tube.
3. Cover the tube with parafilm or use the stopper of the original tube and invert to mix.
4. Centrifuge for 1 minute at 3400 rpm.
5. Discard parafilm into the biohazardous waste.
9. Using a transfer pipet, carefully remove the supernatant and discard it into the sink.
10. Repeat steps 2 through 5 to complete the three washes.
11. Using the automatic pipet, pipet 1.9 ml ( 1,900 ul ) of normal saline and transfer
into Tube#3 labeled “E-5% RCS”.
12. Using another automatic pipette , pipet 0.1 ml ( 100 ul ) of patient’s washed cells ( from
Tube #3 ) into the tube labelled “E-5% RCS”.
13. Cover the tube with a parafilm or use the stopper of the original tube and invert to mix.
This is now your “ Exact 5% washed cell suspension”.
14. Discard all biohazardous waste in a puncture-proof biohazardous waste container.
15. Refer accomplishment of task to your instructor for checking.

C. Preparation of APPROXIMATED 5% Washed Cell Suspension

1. Using a transfer pipette, place 2 drops of “ washed RBC “ ( from Tube #2 ) into the tube
labelled “A-5% RCS”.
2. Gradually add saline solution into the tube and gently mix the tube to re – suspend the
solution.
3. Compare the color of the “A-5% RCS ”to the previously prepared “E-5% RCS” to ensure
that the appropriate strength ( based on approximated color has been achieved. Adjust to
a 3-5% RCS by adding normal saline ( if darker ) or centrifuge to remove excess saline ( if
lighter compared to the “E-5% RCS”.
1. Once the desired color is attained, this is now your “ approximated 5% washed red cell
suspension”
4. Dispose all biohazardous waste into a puncture-proof biohazard container.
5. Refer accomplishment of task to your instructor for checking.
