EXPERIMENT-1

AIM OF THE EXPERIMENT-To prepare a temporary mount of a leaf peel of stomata.

MATERIALS REQUIRED- i)Freshly plucked leaf(Rheo leaf )

ii)Glass slide,

iii)Cover slip,

iv)Petridish,,

v)A pair of forceps

vi)Brush,

vi)Needle.

vii)A pair of sharp scissors

viii)Safranin solution

ix)filter paper

x)Watch glass

xi)Glycerine and blotting paper.

PROCEDURE- i) A freshly plucked Rheo leaf was folded in the centre with the help of
thumb and forefinger in such a way taht the lower part of the leaf remains at the lower side.

ii)One of the two attached parts of the leaf were gently pulled down so that the transparent
leaf peel was seen on the lower side of the leaf.

iii)Small pieces of the transparent epidermis into the petridish filled with water.

iv)A few drops of water was put in a watch glass and the pieces of epidermis was transferred
into the watch glass with the help of brush.

v)A few drops of safranin was added to the watch glass over the pieces of epidermis and was
left for two minutes for the peel to take up the stain.

vi)The peel was transferred back to the petridish containing water which will remove the
extra stain sticking the peel.

vii)A clean glass slide was taken and a drop of glycerine was put on its centre. The piece of
stained peel was transferred on it with the help of a brush.

viii)The cover slip was put over the stained peel gently with the help of a needle.

ix)The excess stain was drain out with the help of blotting paper.
x)The slide was examined first under the lower and then high power of the microscope.

OBSERVATIONS-

i)The epidermis is made up of uniseriate layers of cells.

ii)The cells were closely packed together with distinct cell walls.

iii)The epidermal layer was broken at places ,these lacunae are called stomata.

iv)Each stoma is guarded by a pair of bean shaped cells called guard cells.

v)Guard cells are rich in chloroplasts and one nucleus is present in each guard cell.Inner wall
of guard cell is thicker than its outer wall.

DIAGRAM-

INFERENCE/CONCLUSION- Leaf peel especially from the lower side has many stomata
distributed in it.

PRECAUTIONS- i)The epidermis from the lower side of the leaf should be taken as it is
rich in stomata.

ii)A brush need to be used to transfer the peel to the slide. And peel should never be folded.

iii)A clean slide and cover slip should be used.The cover slip should be placed gently on the
slide to avoid entry of air bubbles.
