Seed Dormancy and Germination

This document describes three laboratory exercises conducted to study seed dormancy and germination: 1. A paper towel method where seeds are placed on a damp paper towel inside a sealed plastic bag to germinate over several days. 2. A water test to determine seed viability by whether they sink or float after 15 minutes in water. 3. A sand germination test where seeds are planted in a tray of moist sand and observed for sprouting over 5 days. Tables track results such as number of seeds germinated and plant growth sizes over time. Questions at the end define key terms and ask about essential seed structures and viability rates.

SEED DORMANCY AND GERMINATION

LABORATORY EXERCISE #3

MATERIALS:
Monggo seeds Reseal able Plastic
Paper towel Short walled container
Water Sand

PAPER TOWEL METHOD

Procedure:
1. Damp the paper towel with water, it shouldn’t be sopping wet but soggy and moist.
2. Put the seeds at the top of the dampen paper towel
3. Put the second paper towel over the top of the seed to make sure the seeds are surrounded in moisture.
4. Then, put the paper towel and the seeds in a sealable plastic bag and seal it.

WATER TEST
Procedure:
1. Put seeds in a water container.
2. Wait for 15 minutes, if the seed sink means they’re viable, if otherwise then the seeds won’t sprout.

SAND GERMINATION TEST
Procedure:
1. Place a 1-2 cm thick layer of washed sand on the bottom of a non-transparent tray.
2. Saturate sand with tap water. The level of the water should not be greater than the thickness of the
sand.
3. Place seeds about 2cm apart in the prepared dish, press the seed gently into growing medium.
4. Use another dish of the same size to cover the seeds, this will increase the humidity for better
germination.
5. Observe the development of the seeds sprouts.

LABORATORY EXERCISE QUESTIONS:
1. Define Germination.

 Germination is the process of a seed starting to grow, or the act of causing a seed to start
growing. It is a development of a plant from a seed after a period of dormancy.
2. Define an abnormal seeds, a dead and a dormant seed.

 Abnormal seeds is a seedling that does not have all essential structures or is damaged, deformed
or decayed that prevents normal development.
 Dead seeds is a seed which are neither hard nor dormant or have not produced any part of a
seedling.
 Dormant seeds means a seed is unable to germinate, even under ideal growing conditions.
3. What parts of the seeds are the essential structures to produce a normal seedling?

 Hypocotyl and Embryo, hypocotyl is a germinating seedling’s stem, normally found underneath
the seed leaves but above the root. Embryo is a diploid, developed from the fertilized egg. All the
cells that need to develop into a mature embryo are present within embryo.
4. What is a good seed viability rate?

 Anything over 50% is decent, some species have lower than 50% viability rate it might be usable
but you may need to plant more seeds to potentially make up for it.
5. In your own opinion, how long do seeds last?

 It depends on the type and the lifespan of seeds. All kinds of seeds have different lifespan. Some
seeds may took a week or a day to sprout, and some will take a year and a half. Plus their viability
rate might also affect in their growth.
