STATEMENT OF PURPOSE

Knowledge is not about width but about depth and research is the way to attain this depth of
knowledge. The thirst of knowing more gives rise to interest on a specific arena and expertise
can be achieved in this way. The each and every step I followed up to this led me to do research
and the next vital step to be taken is to attain an MS degree to explore my interest on a specific
field of research.

“Lab-on-chip Biosensor” a promising field of this century and also challenging to make them
label-free and real time due to their complex fabrication process. The recent advancement on
integrating lab-on-chip with nano-materials and biosensor setup has made it cost-effective and
highly sensitive and due to its diverse application, more research and improvements to existing
methods are required. My cherished dream is to be an accomplished researcher in this arena and
to develop pertinent LOC Biosensor. With that aim in view, I truly believe that pursuing M.S. in
Electrical Engineering from the North Dakota State University will be the ideal one to reach my
dream with its fascinating laboratories to do my research work specially I would like to work in
“Nano-Bio Engineering Lab”.

From my early educational period I was always keen to quest for the explicit view of every
physical phenomenon and consequently, I explore my alacrity to the laws of physics and relating
them to the reality. Engineering, counted as applied physics, could meet my thirst for these
queries and my subsequent academic results supported me to get admitted into Electrical and
Electronic Engineering in “Chittagong University of Engineering and Technology (CUET),
Chittagong”, the premier school of Engineering in Bangladesh. Academically, I always had
excellent records and strived to maintain high standards in all previously attended school, college
and university and always placed among top 5% of my class. I have attained several awards and
scholarships throughout this journey such as Junior talent-pool scholarship from Education
Board, UNESCO scholarship and also university scholarships for all term results. Apart from
academics, I had also been involved in multifarious extracurricular activities like playing sports,
attending in different cultural activities etc., which kindled the spirit of teamwork in me and
enhanced my time management ability. I always placed among top 10% of my class out of 125
students in my undergraduate level and also stood third and first in 6 th and 7th semester
respectively. As a consequence, I secured the 8 th position with CGPA of 3.67 out of 4.00 and
77.40 percent marks. By this time, I tried my best to grasp depth knowledge of EEE courses as
well as laboratory works and projects and also from diverse industrial tours to the substations,
power plant, transformer manufacturing etc. I worked on many small projects using
microcontroller and also attended training on digital circuit design using Verilog, which was
conducted by Avago Technologies, San Jose, USA via online only to few selected students.
Besides those, I attended several courses by a robotic research group and also by online due to
my own interest in enhancing knowledge. As my undergraduate project, I worked on Image
Processing and using MATLAB I developed a new hybrid technique for image compression
which I published in IEEE conference and got citations.

After my graduation, I joined in EEE department of “Faculty of Science Engineering and


Technology” at “University of Science and Technology Chittagong” as a lecturer. Here I have
taught courses and lab related to Electronics, Solid State Devices, Digital Signal Processing,
VLSI etc. and also provided mentorship to project students on image processing, safety blind
stick, perovskite solar cell etc. A thesis work under my mentorship based on ‘Nano structured
perovskite solar cell- material, performance and cost analysis”, has been submitted in IEEE
Conference. As an academic member, I am also involved in student advising, making course
plan for them, arranging seminar and workshop etc. The most amazing thing is to have
prominent colleagues, who are active researchers working in different fields and I have joined
them to attain different skills. From these research groups, I have attained proficiency on
working with OLED (Organic Light Emitting Diode), HEMT (High Electron Mobility
Transistor) and various software skills such as FDTD, COMSOL etc. and have got several
international IEEE publications. These research works have acted as an eye opener to me and
given me the insight of thirst for research. I embarked on the journey by the self-motivated study
on biosensors and worked on design and analysis of nano-structured surface plasmon resonance
sensor. My intention was to improve the performance of this sensor and I found lab-on-chip
applications of biosensors more promising and interesting especially its applications on different
biomedical sectors are being developed and improved day by day.

The recent advances in biosensor have been projected on qualitative and quantitative
determination of various analytes for environmental, clinical, security, food and agricultural
applications. Many of them show excellent performances but still now the real-time operation
and making them label-free is challenging due to the stability of physical transducer and
biological receptor, poor reproducibility of sensors and selectivity in complex matrices.
However, with recent developments in analytical chemistry, there is a possibility for speedy
automation, miniaturization, and integration of a system with high throughput for multiple tasks.
The future lies in the Lab-on-chip biosensor that can accommodate all the essential components
micro-fabricated onto a single chip. Also for the commercialization of such biosensors, more
work is needed to improve the stability, specificity, selectivity, sensitivity, rapid detection, in
vivo measurement, calibration, and ease of production. Due to limited scope of fabrication of
biosensor in my country, I am now only being able to analyze the biosensor performances using
softwares but it is my inherent desire to build compatible biosensor which would be possible
using enriched laboratory like yours.

Both optical and electrochemical immunosensors can be implemented in a microchannel to


facilitate rinsing steps that are labor-intensive and require a skilled operator. In fact, there have
been attempts to provide a higher level of automation and to increase ease of use for
immunosensors (and certainly other biosensors as well), through fabricating more complicated
microchannels. These attempts have usually been achieved by utilizing a concept known as lab-
on-a-chip. Also rationally designed biosensing system supports multiplex analyses is warranted
for medical diagnosis to determine the level of analyte interaction. Recently in a study, the
chemically functionalized novel multi-electrode polysilicon nanogap (PSNG) lab-on-chip is
designed which facilitates multiplex analyses for a single analyte. With the assistance of
microfluidics, Human Chorionic Gonadotropin (hCG) sample was delivered via single-injection
to 3-Aminopropyl (triethoxy)silane (APTES) and Glycidoxypropyl(trimethoxy)silane (GPMS)
modified PSNG electrodes and the transduced signal was used to investigate the dielectric
mechanisms for multiplex analyses. The results from amperometric response and impedance
measurement delivered the scale of interaction between anti-hCG antibody and hCG that
exhibited 6.5 times higher sensitivity for the chemical linker, APTES than GPMS. Under
optimized experimental conditions, APTES and GPMS modified immunosensor has a limit of
detection as 0.56 mIU/ml and 2.93 mIU/ml (at S/N=3), with dissociation constants (Kd) of
5.65±2.5 mIU/ml and 7.28±2.6 mIU/ml, respectively. These results suggest that multiplex
analysis of single target could enhance the accuracy of detection and reliable for real-time
comparative analyses. Their designed PSNG is simple, feasible, requires low sample
consumption and could be applied for any given multiplex analyses. Moreover, a new study
designed a cost-effective, label-free, in situ point-of-care (POC) immunosensor to estimate hCG
using a cuneated 25 nm polysilicon nanogap electrode. A tiny chip with the dimensions of 20.5×
12.5 mm was fabricated using conventional lithography and size expansion techniques and the
sensing surface was functionalized by (3-aminopropyl) triethoxysilane and
quantitatively measured the variations in hCG levels from clinically obtained human urine
samples. From these studies, I have got inspiration to design a new lab-on-chip biosensor which
would be able to perform multiplex analysis as well as label-free and efficient in real time.

Future is always unpredictable, so it’s really hard to say! But I have the confidence to make it in
my way. I always look at what is intellectually challenging, what are the major problems to be
solved in the field of LOC biosensor. I think I am best suited to this program because I like to
perform independent research activities and usually work best in group activities. I feel that with
the comprehensive research facilities and competent faculty members supervising the students,
Electrical and Computer Engineering Department at The North Dakota State University is the
ideal one for me to pursue graduate studies. This program fits with my future plan in every
aspect I can imagine. The fascination for “making something work” and understanding “how it
works”, an aptitude for teaching and teamwork, a zest for challenges and dedication for
innovative research will make my academic aspirations a reality if I am admitted in the M.S
program at your University with financial aid.
