Instructions:

1. This document gives details in apparatus, materials, and procedure for the practical test.

2. Candidate must follow the procedure given and conduct the experiment individually.

3. Candidate needs to show the scientific skills and technical skills during handling the

experiment as well as after conducting the experiment.

4. The data and observation must be record.

5. After completing the experiment, candidates must complete the report within 2 days and

submit the report in soft copy form to class’s lecturer. Late submission will be penalized by

the lecturer.

6. Total marks for practical test are 10% in handling the experiment, 10% in report writing

and 5% for digital skills used.

7. The candidates must follow the safety rules during handling the experiment. Keep your work

area tidy and clean up any spills.

PRACTICAL TEST : BASIC TECHNIQUES IN ISOLATING DNA

Title Isolating DNA


Objective To isolate DNA from plant tissue
Introduction Each chromosome is a single thread-like structure made up of long molecules of
DNA combined with histone protein. The DNA molecule is made up of many small
sections called genes. Shortly before cell division occurs, each DNA molecule
replicates itself. So, one thread of the chromosome becomes two identical
chromatids. As the two chromatids are identical, they will have identical genes.
These identical genes are known as allele. In this experiment, you will rupture fruit
cells, thus releasing their contents such as protein, DNA, RNA, lipids, ribosomes
and various small molecules. DNA is then suspended by alcohol as supernatant
layer.

The purity of DNA will require further steps. After the isolation of nucleic acids, the
solution is still contaminated with proteins which can be removed. To check the
success of the removal, a purity determination is performed, which is based on the
different absorption characteristics of the proteins and the nucleic acids using UV
spectrophotometer.

Procedures 1. Pour 10 ml of ice-chilled alcohol into a boiling tube and place it into a beaker
containing ice cubes.

2. Slice and mash tomato using mortar into fine texture.
3. Transfer mashed fruit into a new beaker.
4. Add 100 ml of the salt-detergent solution. Incubate the mixture in the water
bath at 60’C for 15 minutes.
5. After 15 minutes, sieve the mixture with strainer and collect the liquid in a
clean beaker.
6. Fill in half of the boiling tube with sieved liquid.
7. Very carefully pour 10ml of ice-chilled alcohol into the side of the boiling tube
at flat angle. (Remark: make sure both liquid do not mix and alcohol form a
separate layer on top of the sieved liquid).
8. Observe and draw the extracted DNA between alcohol and the sieved liquid in
the clear container.
