Medicinal Chemistry-1

The document discusses the Structure-Activity Relationship (SAR) of catecholamines and non-catecholamines, highlighting their chemical structures and biological activities. It explains the differences in metabolic stability, receptor affinity, and clinical uses between the two classes of compounds. The conclusion emphasizes that catecholamines have a transient action due to faster metabolism, while non-catecholamines tend to have prolonged effects.

INTRODUCTION

SAR is the Structure-Activity Relationship of any molecule. It will

explains how the chemical structure of a compound affects the
biological activity of the compound. SAR plays a important role in
designing drugs with better efficacy, selectivity, and stability.
CATECHOLAMINES

• Catecholamines is the part of autonomic

nervous system.

• Catecholamines are neurotransmitters and

hormones derived from tyrosine, containing
a catechol nucleus and an amine group.

• Eg - epinephrine, nor epinephrine,

dopamine
SAR OF CATECHOLAMINES

SAR will be studied by-

• Catechol ring substitution
• Substitution at ethylene linkage
• Substitution in amino group

Catecholamines undergoes two types of enzymatic degradation-

• COMT:- methyl group is attached
• MAO:- it will inactive metabolites
NON CATECHOLAMINES

• Non-catecholamines are those adrenergic drugs have lack of

catechol nucleus but still interact with adrenergic receptors.

• Eg- phenylephrine, ephedrine,

amphetamine

Fig:- Structure of non catecholamines

CLASSIFICATION OF NON-CATECHOLAMINES
SAR OF NON CATECHOLAMINES
Structure- activity relsationship of non-catecholamine:-
• Lack of catechol ring
• Increased lipid solubility
• Side chain modification
• Amine group

Non-Catecholamines undergoes two types of enzymatic

degradation-
• MAO:- it will inactive metabolites
• Other liver enzymes
DIFFERENCES

Catecholamines Non catecholamines

CONCLUSION
Catecholamine and non-catecholamine differ significantly in their
chemical structure, physiological effects, and pharmacological
properties. The difference between catecholamine and non-
catecholamine involve their metabolic stability, receptor affinity, and
clinical uses, with catecholamine generally being more transient in
action due to faster metabolism, while non catecholamine tend to
prolonged effects due to their resistance to break down.
