Make gut extract:


Surgical carp on the stomach.
Separate the gut of other organs carefully. Take the small intestine
by cutting from the latter part of the stomach to the beginning of
the large intestine.
Take the bile bag carefully and not to break
Open small intestine by means of longitudinal slashed.
Clean the small intestine of the premises of distilled water, and
then insert it into the mortar.
Take 20 ml of 50% glycerol and insert it into the mortar, and puree
intestines. Take 4-5 drops toulen, puree back. After the intestinal
smooth enter into the bottle, then close tightly. Wrap the bottle with
carbon paper.
Save the gut extract in the dark for 6-7 days.
After 6-7 days, strain the gut extract with filter paper.
Perform the test on the filtered solution is proving tests amylase,
maltase, and trypsin.

b. Test effect of bile for fat:


1. Prepare 2 Test Tube,
2. Give Label A And B
3. Take in bile into test tube a with little cut in surface
4. Dilute the bile with aquades become 20 ml
5. Take in 20 ml aquades into test tube b as control
6. Add to each test tube 2 ml cooking oil
7. Shake it and allow it to 5-10 minutes
8. Observe what happened at 2 substance that in test tube
9. Compare how big the clots of fat in each test tube

c. Tests proving the existence of amylase:


1. Provide two test tubes and labeled A and B. Pour benedict reagent
into the tube 2 ml respectively.
2. Prepare sua another tube and labeled C and D.
3. Enter cooked starch solution dilute each 2 ml into the tube C and D.
For C tube add 1 ml of extract specific whereas for D tube add 1 ml of
distilled water. Shake the tube stretcher for 5-10 minutes.
4. Apply a drop of as much as 5 drops of the solution in the tube C to
tube A, and the solution in the tube D to tube B.
5. Heat the tubes A and B for 5 minutes and observe the color change
that occurs in the tube solution A and B.

d. Tests proving the existence of maltase:


1. Provide two test tubes and labeled A and B. Pour benedict reagent into
the tube 2 ml respectively.
2. Prepare sua another tube and labeled C and D.
3. Enter maltose dilute each 2 ml into the tube C and D. For C tube add 1
ml of extract specific whereas for D tube add 1 ml of distilled water.
Shake the tube stretcher for 5-10 minutes.
4. Apply a drop of as much as 5 drops of the solution in the tube C to tube
A, and the solution in the tube D to tube B.
5. Heat the tubes A and B for 5 minutes and observe the color change
that occurs in the tube solution A and B.

e. Test proving tripsin


1. Prepare 2 test tube
2. Give the label a and b
3. Add to a and b 1 ml white egg dilute
4. Burn in after boiling
5. Chill its tube
6. After it add the extract of intestine to test tube a 1 ml
7. Add 1 ml aquades into test tube b
8. Chill it 5-10 minutes
9. Drop reagen biuret 5 drops to test tube a
10. Observe the change of color which happened in test tube
