Basic Skill - Prepare Solution of Known Concentration

This document provides instructions for preparing solutions of known concentration and reagents. It outlines the uses of such solutions, which include titrations, instrument calibration, method validation, and staff training. The document discusses calculating concentration, selecting suitable materials, practical preparation steps, labeling, and storage of solutions. It provides examples of how to prepare specific standard solutions and reagents from stock solutions of known concentration.

Outline
What are known concentration & reagent solutions ???
Known concentration solutions: the concentration of
the solution is known with a high degree of certainty
Reagent solutions: aren’t involved directly in the
determination of the concentration of the analyte in test
samples, their concentrations do not generally need to be
known with the same degree of certainty as standard
solutions

PREPARING SOLUTIONS OF KNOWN CONCENTRATION

Uses of solutions of known concentration

In titrations

To calibrate an instrument

To evaluate the performance of test methods both during

method validation and in on-going quality control when
the method is in routine use.
To train staff and evaluate their performance.
Uses of solutions of known concentration
Terms used to describe solutions of known concentration
Uses of reagent solutions
 Many different types of reagent solution used in the laboratory
– Indicator solutions
– Buffer solutions
– Solvents…

3. Calculation
Suggest how to prepare the following solutions:
1. 100 mL of a 0.1 mol L-1 solution of HCl acid from a stock solution with a
concentration of 1 mol L-1.
2. You have a protein solution (a stock solution) at 2 mg/ml. You need to make the
following levels: 400 μg /ml, 100 μg /ml, 20 μg /ml, 5 μg /ml and 1 μg /ml. You
need 50 ml of each concentration.

3. Calculation

The 5 μg /ml and the 1 μg /ml solutions have large dilutions and
volumes of the stock too small to pipet accurately ???

6. Labelling and storage
 Solutions should not be stored in volumetric flasks, transfer them to a suitable
container for storage.
 Ensure that solutions are stored correctly (in refrigerator, amber bottles…)
 All solutions should be clearly labelled with the following information: (the name,
date of preparation, analyst, review or expiry date, hazard information …)
 The label must be securely attached to the container and be written in water
insoluble ink.
