Preventive Maintenance Procedure for Access & Access 2

Latest Revisions Release of the Lean PM Procedure. Due to the large number of changes in this
version they are unlisted.

Purpose This document provides the procedures and information needed to perform the
Preventative Maintenance (PM) procedure for the Access and Access 2
Instruments.

Warning Wear a lab coat, protective gloves and safety glasses when
necessary.

PM Intervals These are the PM intervals and the estimated times for completing Minor and
Major PMs.

PM Checklist Launch the attached PM Checklist form. Select Yes to open as a Read Only
document. The cursor will be in the first form field. Enter the information as
requested, and press the Tab key to advance to the next form field. When all
form fields are completed, select "Save As" from the File menu to save the
document, with the desired file name, to the desired location on your computer.

Initial Evaluation Perform these steps to review the instrument event logs and other information
that may be important to the PM.

Step Action
1 Print the alignment report for future reference.
2 Review the Event Log for any unusual performance issues and note
any issues to address during the PM on the checklist.
3 Review the last several system check results. Note any issues to
address during the PM on the checklist.
4 Review the QC for any unusual performance issues and note any
issues to address during the PM on the checklist.
5 Review the operator's log, if available, and ask the operator to
describe any concerns or issues with the instrument. Note any issues
to address during the PM on the checklist.

Instrument Perform these steps to prepare the instrument for the PM procedure.
Preparation

Step Action
1 Ensure that all appropriate modifications are completed.

z Refer to Oracle for a list of open MODs for the instrument.


z For a listing of modification instructions refer to Chapter 9 of
the Access Service Manual.
z If there is time to upgrade the system and there are parts
available, then install them at the appropriate step in the
procedure.
2 Verify the reagent storage temperature using a thermocouple.
o
If the temperature does not agree with the on screen display within 2
C, then replace the Reagent Storage thermistor.
3 Verify that the temperatures displayed in the User Interface and their
associated duty cycles are within the appropriate range.
Duty cycles greater than 0.75 should be investigated.
4 Unload any onboard reagent packs and place them in a refrigerator or
cooler.
5 For a PM Major Only:
Note any environmental issues to address with the customer. These
could include instrument clearances, the temperature of the lab, etc.
6 Remove the top front and bottom front covers and bypass the
interlock.
7 Verify that all cooling fans are operating and replace as needed.
z 3 Card cage
z 1 Right side panel
z 1 Power supply (fan not serviceable, replace power sled)
z 1 Peltier

8 Power down the instrument and inspect the card cage for excessive
dust build-up.

Clean the dust out of the card cage only if necessary.


Otherwise, defer cleaning for a subsequent visit.

Sample/Reagent Perform these steps to service the Sample/Reagent Carousel Module.


Carousel Module

Step Action
1 For a PM Major Only:
Remove the Sample / Reagent Carousel Module from the instrument
and clean under the Module.
Remove any dust from the instrument exhaust fan located on the right
hand side of the Sample / Reagent Carousel Module.
2 Remove the Reagent Storage module cover, then clean the underside
of the cover.
3 Remove and clean the sliding door on the cover using a damp lint free
tissue.
Lift the right end of the door (enough to clear the end stop)
and slide it to the right until it is completely out of the slots.
4 Clean the aspiration slot and sliding door channel using a damp lint
free tissue or swab.
5 Re-install the sliding door, with the magnet to the left, and ensure that
the sliding door operates smoothly.
6 Remove the Reagent Storage Wheel, clean any residue from the
reagent nests. Clean the interior of the Reagent Storage Module.
7 For a PM Major Only:
Reassemble the Sample / Reagent Carousel Module and install it in
the instrument.

Analytical Module Perform these steps to service the Analytical Module Incubator Belt and
Incubator Belt Pulley Bearings.

Step Action
1 Remove the Analytical Module from the instrument. Refer to the
"Analytical Module, Removal / Installation" procedure for details.
2 Remove all RVs from the waste chute. (remove waste bag)
3 Remove the plastic module cover and the light shield.
4 For a PM Major Only:
Remove and discard all vessel holders.
5 Loosen the left-front incubator belt idler pulley, then remove the
incubator belt. Refer to the "Belt, Incubator Replacement" procedure
for details.
z Inspect the incubator belt for cuts or cracks and discard if any
are noted.

z Do NOT replace the incubator belt during a PM unless


damaged.
6 Exceeding 0.030" (0.76 mm) may indicate bearing failure.
Ensure that the incubator belt pulleys rotate smoothly in both
directions, and that vertical play in the pulleys is not excessive
7 Inspect the Analytical Module and identify the locations of any spills or
residue, including any on the light shield, and perform corrective
action to prevent future leakage.
8 Clean the incubator track as necessary.

Mixer / Wash Arm Perform these steps to service the mixer area components.
Area

Step Action
1 Remove the Mixer Motor / Belt tensioner assembly to free mixer belt
from the pulleys.
Remove and discard the Mixer Belt.
2 Remove the 3 mixer pulleys from the Analytical Module.
z If performing a Major PM, then discard the mixer pulleys.
z If performing a Minor PM, then clean the mixer pulleys with
deionized water and inspect.

z If the mixer pulleys are difficult to remove or are covered with


deposits, then apply deionized water to them before
attempting removal.
z If the plastic lower snap-in bushing does not come out of the
module with the pulleys, then thread the long light shield
mounting screw into the bushing and lift upward to remove.
3 Install the 3 mixer pulleys.
4 Remove the 3 mixer pinch roller assemblies.
z If performing a Major PM, then discard the pinch roller
assemblies.
z If performing a Minor PM, then clean and inspect the pinch
rollers for wear and spring to housing fit.
5 Install the 3 mixer pinch roller assemblies.
6 Move the Wash Arm linear slide up and down the lead screw. The
movement should be smooth without binding between the nut and the
lead screw. Also, the Wash Arm Linear Slide should not be loose
(wiggle) when moved.
7 Remove the two screws holding the linear nut band to the linear slide
and then check the lead screw mountings/bearings for play.The lead
screw shaft should not move up/down in the bearings more than .010"
(0.25 mm). Replace if necessary. Refer to the "Wash Arm, Lead
Screw Replacement" procedure for details.
Excessive up/down or side to side looseness could indicate
bearing failure.

8 Do not lubricate the lead screw, the plastic linear nut is


self lubricating.
Lightly lubricate the wash arm linear slide with type 2 lithium grease.
9 Replace the Mixer Belt and install the Mixer Motor / Belt Tensioner
assembly.
Ensure that the mixer belt is routed correctly over all the
pulleys.
10 Reattach the linear nut band to the linear slide.

Wash / Read Area Perform these steps to service the Wash Carousel and Luminometer.

Step Action
1 Remove the Wash/Read Carousel insulating cover, the Wash/Read
Carousel cover plate, and the Wash/Read Carousel.
2 Clean any dried deposits off of the wash/read carousel.
The Wash/Read carousel may be soaked in hot water to
soften deposits.
3 Rotate and inspect the 6 wash carousel bearings. Replace any
defective bearings.
4 Inspect the Luminometer lens. If the lens is dirty, then carefully clean
with a non-cotton tipped applicator.

z Do not blow with canned air, it may dislodge the Luminometer


lens.
z Protect the Luminometer from bright light.
5 If the spring is damaged, flat, or corroded, then replace it.
Inspect and clean the RV retainer spring assembly.
6 Reinstall the Wash/Read Carousel, the Wash/Read Carousel cover
plate, and the Wash/Read Carousel insulating cover.

Install Incubator Perform these steps to install the Incubator Belt.


Belt

Step Action
1 Install and tension the incubator belt. Refer to the "Tool, Tensioning,
Incubator Belt" procedure for details.
2 Install new vessel holders, and a new home position vessel holder
(with magnet on top).
3 Do not reinstall the Analytical Module yet. This is done in
a later step.
Verify that the belt guides are just flush with the incubator belt. Adjust
any that are not aligned properly.

Fluidics Module Perform these steps to service the Fluidics Module.


Use paper towels where needed to absorb any fluid drips or leaks
from the probes and the main pipettor while performing these steps.

Step Action
1 Inspect all fluidics tubing and fittings for leaks or damage.
2 Inspect and clean the Substrate Probe with Citranox. If the substrate
probe is kinked or damaged, then replace it.
For a PM major only: Replace the Substrate Probe.
3 Remove and replace the Aspirate Probes (discard the old ones).

z Clean the new probes before installation.


z PM kits contain only one set of aspirate probes. Subsequent
sets must be ordered by the customer as a consumable.
z Use the Aspirate probe brushes to clean the inside of the
replacement Aspirate probes.
4 Clean the Dispense Probes with deionized water and a lint free nylon
swab.
5 Wipe any debris off the Peri Pump rollers and the
cartridges and ensure the cartridges are put back in the correct order.
Trim to fit and then install new peristaltic pump tubing. (3 small, 2
large tubes)
6
z Place the pipettor probe and dispense probes over absorbent
material to absorb any draining buffer in these steps.
z Do not lose the pump to manifold o-ring during this step.
For a PM major only:
Remove the Wash Pump and:
z clean the pump
z install a new seal and o-ring
z If the belt is loose/stretched or frayed, then replace it.

7 Do not lose the pump to manifold o-ring during this step.


Remove the Precision Pump and:
z clean the pump
z If the belt is loose/stretched or frayed, then replace it.
z inspect the precision pump spring.
z install new seals and o-rings
The upper seal is smaller than the lower seal and
has a small groove on its circumference.
8 Disassemble, clean, inspect, and reassemble the Wash and Precision
valves. Refer to the "Valves Ceramic, Cleaning" procedure for
details.

Pipettor Module Perform these steps to service the Pipettor Module.

Step Action

1 Inspect the wash tower area for leaks or salt deposits. Clean as
necessary.
If performing a major PM, then install a new wash tower nozzle and
o-rings.
2 Do not use the Pipettor torque wrench to remove the probe.
Remove the main pipettor probe using 5/16" and 1/4" open end
wrenches and install and torque the new pipettor probe.
3 Lightly lubricate the linear slide on the pipettor carriage assembly with
type 2 lithium grease.
Clean the upper and lower crossbars with a lint-free cloth, then apply
a light coating of type 2 lithium grease only to the upper crossbar.
4 Check belt tensions and mounting clips for looseness.
5 Visually inspect the service loop and replace if cracked or worn.
Verify that the Pipettor Assembly moves smoothly through its full
range of travel.
6 Clean the instrument baseplate, fluidics manifold area, power sled
intake fan, left side wall and fluidics tray area.

Power On Perform these steps to power on the system.

Step Action
1 Install the Analytical Module.
2 Mount the Wash Arm and check that all cables and tubes are
attached.
3 Visually inspect the incubator belt, RV shuttle, and wash/read
carousel paths to ensure that none will interfere with each other during
initialization.
4 Power on the instrument and initialize.
The Incubator Belt, RV Shuttle and Wash/Read Carousel
may need to be homed in diagnostics for initialization to begin.

Pipettor Alignment Check and adjust the Pipettor alignments by referring to one of these "Pipettor,
Alignments / Adjustments" procedures:
z Access 2
z Access base

Incubator Belt Perform these steps to calibrate and align the Incubator Belt and perform the
Alignment & exerciser routines.
Exercisers

Step Action
1 Calibrate the Incubator Belt. Refer to the "Belt, Incubator, Calibration"
procedure for details.

2 Check and align the Incubator Belt at the home, shuttle transfer,
wash-in, and wash-out positions. Refer to the "Belt, Incubator,
Alignments" procedure for details.
3 Load RVs and perform 2 cycles of the vessel holder exerciser test.
Refer to the "Theory of Operation Exercisers" procedure for details.
4 Perform 2 cycles of the wash carousel transfer exerciser test and 2
cycles of the incubator track exerciser.
Refer to the "Theory of Operation Exercisers" procedure for details.

Aspirate Probe Perform these steps to align the Aspirate Probes.


Alignment

Step Action
1 Verify that all probes are aligned in the center of the holes in the
wash/read carousel housing.
If they are not centered, then carefully bend the probes to center
them.
2 Verify that the Aspirate Probes move upward approximately 0.070"
(1.8 mm) when pulled, and spring back down when released.
3 Verify the pinch roller bracket adjustment and the Aspirate Probe Z
alignment. Refer to the "Wash Arm / Probe Plate Adjustment"
procedure for details.

RV Sensor Voltage Perform these steps to check and adjust the RV Sensor Voltages.
Adjustment

Step Action
1 Clean the RV sensors with a non-cotton tipped applicator and
deionized water.
To access the RV sensors, rotate the slotted shaft in front of
the RV shuttle motor to move the walking beam to a location where
the sensors are visible.
2 Check the voltages of the RV sensors at VS1 and VS2 on the
Analytical interface PCB and adjust as necessary. Refer to the
"Sensors, RV Adjustment" procedure for details.

Ultrasonics Perform these steps to verify the Ultrasonic Transducer operation.


Adjustment

Step Action
1 Verify that the Ultrasonic Transducer temperature is within
specification using a thermocouple. The Transducer temperature
o o
specification is 35 C to 39 C.
2 Check and adjust the Ultrasonics Voltage and Frequency. Refer to the
"Ultrasonic PCB, Adjustment" procedure .

Mixer Speed Verify that the mixer speed is 2500 RPM +/- 20 RPM in both CW and CCW
Adjustment directions by referring to the "Motor, Mixer, Speed Adjustment" procedure .

Vacuum System Perform these steps to verify proper operation of the vacuum system.

Step Action
1 The presence of more than 1/8" fluid in the bottle may
indicate a problem with the large peri pump tubing, a clogged air filter,
or restriction in the waste drain tubing.
Drain the vacuum bottle if it contains more than 1/8" (3 mm) of fluid.
2 Flush the vacuum pump. Refer to the "Pump, Vacuum, Flushing"
procedure for details.
3 The Air filter bottle may become clogged with dried buffer
residue causing slow vacuum increase or limiting maximum vacuum.
Replace the air filter if it blocks the air discharge from the vacuum
pump.
Operate the vacuum pump for one minute, then activate the vacuum
valve. Verify that the vacuum pressure operates as expected.
These are the expected Vacuum values:
z 0 +/- 10 mmHg (vacuum pump off).
z 30 to 60 mmHg (vacuum pump on and vacuum valve off).
z Greater than 450 mmHg within 30 seconds (vacuum pump on
and vacuum valve on).
z Less than 50 mmHg leakage in 30 seconds (after pump is
powered off with valve on).
z 0 +/- 10 mmHg within 2 seconds of (vacuum pump off and
vacuum valve off).

Dispense And Perform these steps to verify proper operation of the Dispense and Aspirate
Aspirate Systems systems.

Step Action
1 Prime the fluidics and check for any leaks.
2 If fluid is not being aspirated or dispensed, press [Cancel]
and perform the appropriate corrective action(s).
During the priming sequence, verify that fluid is dispensed by all 3
Dispense Probes and is aspirated by all 3 Aspirate Probes.

z During aspiration, verify that each of the 3 Aspirate Probes
raises up when touching the bottom of the RV and is not
pulled upward by its tubing.
z If the instrument is on software version 3.30 or 2.5 and higher
and Original Wash Buffer, then the RV should contain less
than 5 µL of fluid. Aspirate volume checks performed on any
other configuration should result in less than 4µL of fluid in the
RV.

Perform the Aspirate volume check.


4 Perform the Substrate Volume check and inspect the 3 RVs for
bubbles.
5 Perform the Dispense Volume check for each probe and inspect the 3
RVs for bubbles.

z For software versions below 3.30 (Access Base) and 2.5


(Access 2), if bubbles are present, then adjust the Wash
Pump Backlash. Refer to the "Pump, Wash, Backlash
Adjustment" procedure for details.
z The Wash Pump Backlash is no longer manually adjusted in
software versions 3.30 and 2.5 and above. The adjustment is
automatic.
6 Perform the Pipettor Volume check for 50 µL and 200 µL.
7 Calibrate the Obstruction Detection feature for Access 2 if installed.

Verification Perform these steps to verify proper operation of the instrument.

Step Action
1 Dispense probe 1 should be removed when reinstalling
the light shield.
Reinstall the light shield, plastic incubator cover and the instrument
covers and close all covers.
2 Print an alignment report and an incubator belt calibration report and
leave them with the customer.
3 Do not leave the interlock bypass key at the customer
site.
Ensure that the cover interlock bypass key is removed from the front
cover interlock switch and ensure that the utility assay is enabled.
4 Use original System Check solution. The use of HS
System Check solution will cause Washed %CV failure.
Perform the system check procedure and correct any problems
identified.
5 Review the DCF value shown on the last several system check
results.
If the DCF is not between 0.90 and 1.10, then adjust and save the
high voltage (HV) control value. (Refer to the "Luminometer Assembly
Adjustment" procedure for details.)
6 Complete any additional testing according to the "Verification of
Service Repair" document .
7 Inform the customer of their need to run all levels of QC prior to
reporting patient results.
Recalibration may be needed to correct QC shifts following a
PM.
8 Reload reagents and start QC. Ensure the first nine results are within
2 standard deviations of the mean.

9 Clean the work area and dispose of used parts/materials according to


the facility's guidelines.
10 Complete the PM Checklist and file any PM documents according to
procedure.
11 Leave a signed copy of the PM Checklist for the instrument
maintenance record book.
