II. PROCEDURE:

A. POTATO OXIDASE
1. Prepare three clean test tubes. Label test tubes 1 to 3.
2. Pipette 1 mL of catechol solution and small amount of potato extract in tube
1.
3. Pipette 1mL water and a small amount of potato extract in tube 2.
4. Pipette 1 mL of catechol solution and 1 mL water in tube 3.
5. Mix the contents of the tube by shaking gently and place in a water bath at 37
deg. C.
6. Examine the tubes after 5 minutes by holding up to light. Record the results.

Result (5 points):In tube 1, the color turned dark brown due to the formation
of benzoquinone in the presence of potato oxidase enzyme. So the reaction
happened successfully. In tube 2, there was no substrate so no reaction
happened. In tube 3, the reaction did not take place since there was no enzyme
to help get the necessary activation energy for the reaction to happen. Hence
its proved that enzyme is needed for the reaction to take place efficiently.
B. CATALASE

1. Prepare 2 clean test tubes. Put a small slice of cooked potato in one tube, and a
small slice of fresh potato in the other.
2. Add a few drops of hydrogen peroxide. Observe the rate of reaction and record
the results.

Results (5 Points): The tube with the cooked potato does not contains the
enzyme as cooking has denatured the enzyme so no reaction. In the tube with
the fresh potato, we can notice that the catalase enzyme present in the potato
reacts with hydrogen peroxide to produce water and oxygen. Hence the
presence of enzyme has caused the reaction.

III. QUESTIONS: (5 points each question)

1. Why should peeled potatoes not be exposed to air longer than necessary?
When peeled potatoes are exposed to air, the color of exposed surface
changes. The brown color is a result of the catalyzed by several enzymes like
polyphenol oxidase, monophenol oxidase and cytochrome oxidase.

2. Compare the stability of oxidase and peroxidase to heat.


Peroxide is a very heat stable enzyme so that heat is used frequently as an
index in the effectiveness of blanching, the enzyme is held at 85°c, half of the
original activity remains it is known to self-regenerate due to reversible
denaturation. Oxidase is not stable at this temperature even with the addition
of catechol.

3. Differentiate the action of oxidase, peroxidase and catalase.


A. Oxidase:
-Polyphenol oxidase is an oxygen transferring enzyme. It activates molecular oxygen and
enhances oxidation. It catalases oxidation involving molecular oxygen. Oxygen as e- acceptor
and reduces to H²O and H²O².

B. peroxidase:
-It is well suited for preparation of enzyme conjugated antibodies, due in part to its ability to
yield chromogenic products, and in part to its relatively good stability characteristics .

C. Catalase:
-It is an antioxidant enzyme produced in the body. It converts hydrogen peroxide into
hydrogen and oxygen, preventing the formation of carbon dioxide bubbles in blood.  It also
uses H2O2 to break down potentially harmful toxins in the body, including alcohol, phenol
and formaldehyde.
