Always refer to the system manual when working near locations


at which the alert mark shown on the left is attached. If the
operation, etc., is performed without heeding the advice in the

When supplying power to this equipment, connect the


accessory 3-pin power cord to a 3-pin grounded power outlet. If
a grounded 3-pin outlet is not available, use a conversion
adapter and ground the green wire. If power is supplied without
grounding the equipment, there is a risk of receiving a fatal
electric shock and equipment damage.

Before changing the fuses, ALWAYS remove the power cord

Clean the power line regularly; if dust accumulates around the
power pins, there is a risk of fire.
Keep the equipment clean so that the ventilation holds are not

Periodically clean your equipment's case using a damp cloth.
Do not use abrasives, cleaning solvents or strong detergents, as
they may damage the finish or affect the reliability of the
structural components.

I. Pre-Installation

I-1. Introduction
Lumina fluorescence spectrometer is designed to provide a complete analysis system
for fluorescence, phosphorescence and chemiluminescence measurements.
All instruments will be tested and proven to the performance before the shipment

Preparation Done

The installation place should be prepared / checked. □

The electric power is available / stable. □

The system and accessories should be unpacked on site. □

Proper computer system should be prepared. □

Windows operating system should be prepared. □

Quartz cell for fluorescence is available. □

Operation training is required. □

I-2-2. Environmental Conditions

You are responsible for providing an acceptable operating environment. Attention paid
to the operating environment well ensures the continued high performance of your
Lumina fluorescence instrument. The instrument warranty will be made void if the
equipment is operated under sub-standard conditions.

Humidity

The relative humidity of the operating environment should be lower than Maximum
75% with no condensation. Operating the Lumina fluorescence in very low humidity
may result in the accumulation and discharge of static electricity that shortens the
life of electronic components. Operating at high humidity will produce condensation
and result in short circuits. Thermo Scientific recommends that your laboratory be
equipped with a temperature / humidity monitor. This will ensure that your laboratory
is always in conformance with the temperature and humidity specifications.

Temperature

Air conditioning is recommended. The room should be temperature controlled if


your analyses are particularly sensitive. For optimum analytical performance it is
recommended that the ambient temperature of the laboratory be between
15-35℃ and be held constant to within +/-2% throughout the entire working day.

As laboratory temperature increases, system reliability decreases and all


electronic components generate heat while operating. This heat must be
dissipated to the surrounding air so that the components are operating correctly.

Vibration

Ensure that lab benches are free from vibration. If possible, any equipment
generating vibration during the operation should be installed on the floor rather than
alongside the system on the workbench.

Work bench

The workbench must be stable and durable enough to support the total weight of
equipment. The worktops should be large enough to permit a free circulation of air
around each unit in the system. Remember to provide space for the computer,
monitor and printer. To avoid damage through spillage of the samples being
analyzed, the worktops should be covered with a material which is corrosion
resistant and impervious to liquids.

I-2-3. Electrical Specifications

In view of the electronic components in the Lumina system, it


should be positioned well away from any instruments emitting an
electromagnetic field and any environment that may cause
vibration.

Power supply

All power supplies must be single phase AC voltage, three wire system (active,
neutral, ground) and should be terminated at an appropriate power outlet and/or
regulations imposed by local authorities responsible for the supply of electrical energy
to the workplace.

A grounded AC power supply must be used. The Lumina accepts a dual voltage of

either 110 or 220V at 50 ㎐ or 60 ㎐ and requires 300 W power including computer

and printer.

Good electrical grounding is essential to avoid potentially


hazardous shock hazards. 3-wire outlet with ground connection
must be provided for the instrument. Make certain that power
outlets are grounded at the grounding pin.

Avoid using power supplies from a source that may be subject to electrical or RF
interference from other services (large electric motors, elevators, and welders for
example).

Two fuses are used in the Lumina fluorescence spectrometer;


Fuse A: AC 250V, T2AL
Fuse B: AC 250V, T5AL

I-2-4. Equipment on-site

Insurance

As the carrier’s liability ceases when equipment is delivered, Thermo Scientific


recommends that the instrument owner arrange a separate insurance to cover
transportation from the delivery point to the installation site. The delivery point will vary
according to the carrier, the shipping method and in some cases the terms of sale. Some
carriers will deliver only to their own distribution centers, who then will deliver to the
actual installation site.

Inspection for transit damage

Transit damage can be obvious or concealed and in either case will be admitted by the
carrier only if it is reported within the terms of the carrier’s agreement. For any claims
against damage in transit, the following general rules apply;

- Before accepting delivery, you must inspect the packages for signs of obvious
damage. The nature of any obvious damage must be noted on the carrier’s waybill,
which then must be countersigned by a representative of the carrier

- Within the time limit stated in the terms and conditions of carriage, a further inspection
must be made for concealed damage. If any damage is found at this stage, the carrier
must be modified in writing. You must retain all packing material for subsequent
inspection by a representative of the carrier.

- A copy of any damage report must be forwarded to Thermo Scientific or Sales


representative

- ‘SHOCK WATCH vibration indicator label that helps you to aware of the damage level
are attached on Packing Boxes. If the indicator color is changed as red color, it means
the packing box could be impacted/damaged by the physical shock externally.
If it is happened, contact Thermo Scientific or sales representative immediately to hear
the next procedures.

II. Xenon Lamp Replacement

II-1. Xenon Lamp Replacement


The Xenon lamp for replacement is packed in the manufacturer’s box and must be
installed in the lamp housing correctly. Read all the precautions before attempting to
replace the Xenon lamp.

Caution

A. Xenon arc lamp has an explosion hazard. Make sure the system’s power is off
and all AC power is disconnected from the system. Read and follow the cautions
presented below.

B. Xenon arc lamp has an explosion hazard. Wear goggles and protective clothing
when opening the lamp housing and when handling the lamp.

C. The lamp will remain extremely hot for approximately one-half hour after it has
been turned off. Do not touch the lamp or the metal unit until the lamp has cooled.

D. Never look directly at the xenon arc or its reflection. Intense radiation can
permanently damage eyes.

E. Do not touch the focusing lens, back scatter mirror, or the surface of the lamp.
Fingerprints will be burned onto these surfaces when the lamp is ignited.

2) Turn off the power of the instrument.
3) Remove white screw caps (total 10) located around on the upper case.
4) Remove Phillips Head screws using a Phillips screwdriver.
5) Remove Phillips head screws (4 ea) located on the Sample Cell Assay part using a
Phillips screwdriver.
6) Separate the Sample Cell Assay part and lift the upper case.
7) Get rid of socket screws (4 ea) using a hexa socket screwdriver.
8) Separate the lamp connector from the lamp power module and push the hap and pull
out the connector.
9) Remove the lamp mounting module from the lamp housing.
10) Remove the screws (2 ea) located on the edges of the lamp.
11) Unfasten the socket screws (2ea) located on the (+) side of the Teflon bracket.
12) Unpack a new Xenon lamp.
13) Install the lamp in the correct polarity.
14) Fasten the socket screw (2ea) located in (+) direction Teflon bracket. Put the changed
lamp module back in the lamp housing.
15) Finally, connect the lamp cable to the power supply cable.

13. Do not use any tools to tighten nuts when fixing the lamp, use hands only. Otherwise it
could cause breakage.

VI. PMT Replacement
VI-1. PMT (Photo Multiplier Tubes) Replacement
A high voltage is applied to PMT during operation. Always provide adequate safety
measures to prevent the operator or service personnel from electrical shock and the
equipment from being damaged.

Caution
A. Handle tubes with extreme care.
PMT has evacuated glass envelopes, allowing the glass to be scratched or subjected to
shock can cause cracks. Take extreme care during handling, particularly for tubes with
graded sealing on synthetic silica bulbs.

B. Keep faceplate and base clean.


Do not touch the faceplate and base with bare hands. Dirt and grime on the faceplate
causes loss of transmittance and dirt or grime on the base may cause ohmic leakage. If
the faceplate becomes soiled, wipe it clean using alcohol.

C. Do not expose to strong light.


The PMT may be damaged if exposed to direct sunlight or intense illumination. Never
allow strong light to strike the photocathode.

D. Carefully handle tubes with a glass base.


PMT with a glass base (also called button stem) are less rugged than tubes with a
plastic base, so sufficient care must be taken when handling this type of tube. When
fabricating a voltage-divider circuit by soldering resistors and capacitors to socket lugs,
solder them while the tube is fully inserted into the socket.

E. Helium permeation through silica bulb.


Helium will permeate through silica bulbs and increase noise, leading to damage that
makes PMT unusable. Avoid operation or storing them in and atmosphere where helium
is present

VII-1. Monochromator Replacement


Monochromator is dispersing polychromatic or white light into the various colors or
wavelengths. This dispersion can be accomplished using diffraction gratings.
The performance specifications of a monochromator include the dispersion, the efficiency,
and the stray light levels. It is important to recognize that transmission efficiency of a
grating and monochromator is dependent on wavelength and on the design of the grating.
Moncochromator consists of a grating, a step motor, reflect mirrors, a worm gear, a worm
wheel, and photo interrupt sensors.

[Power supply]
The power supply of the xenon lamp is extremely dangerous, it generates the high
voltages so take a great care to the electric shock.

[Xenon lamp]
Xenon lamps emit ultraviolet radiation which is harmful to the eyes and the skin so
Always wear a light-shielding protector while the lamp is on.

[Grating]
1) Never touch grating surface. Crusts do not affect on optical characteristics in most
cases but fingerprints cause surface deterioration.
2) Do not wipe the sides of grating by solvent and /or clip the edges of grating plane.
When liquids and/or oil ingredients are adhered in the area of the edges of grating

plane, they might intrude into grating plane alongside of grooves and diffuse over
grating plane.
3) Do not rinse out gratings by water and/or solvent. Also, be sure not to expose
gratings to chemicals and/ or that vapor which might exert harmful influences on
resin and/or coating materials of gratings. They cause surface deterioration.
4) Use gratings in low dust environment. Do not breathe upon grating surface when
dust and/or dirt are accreted. Gently blow away by dry and clean air.
5) Use gratings in no-dew-condensation environment. Dew condensation causes
eminent deterioration of optical characteristics.

[Air Fan]
Disconnect the cable of air blow fan for protection of grating during the service.
Oil of DC motor could attack the surface of grating.

16. 1) Place the upper optic module and screw socket head screw. (Refer to below
figure.)
2) Open the shutter.

Do not touch the faceplate and base with bare hands. Dirt and grime on the
faceplate causes loss of transmittance and dirt or grime on the base may cause
ohmic leakage. If the faceplate becomes soiled, wipe it clean using alcohol.

10. Put a new emission monochromator into the base plate. Reconnect all the cables of
the worm drive step motor, the filter step motor and the slit step. All the cables should
be connected in their proper positions.

11. Fasten the socket head screws (total 4ea). Pleas don’t fasten screws tightly since
Monochromator need to be moved slightly for beam Alignment.

12. Run the ‘Alignment.exe’ for alignment procedure located in Luminous Software folder

13. Turn on the xenon lamp, and Calibrate Slits and Filters using Alignment software.
After finishing calibration of slits and filters, set the filter as ‘air blank’ and the slit 2.5nm
and move the Excitation monochromator grating to zeroth order for beam alignment.

Xenon lamps emit ultraviolet radiation which is harmful to the eyes and the skin so
always wear a light-shielding protector while the lamp is on.

14. Install Teflon block in the cell holder. The Teflon block (PN FS2-ACCE-08) reflects the
xenon lamp beam from the excitation lens holder to the emission lens holder and that
will be helpful to make an emission monochromator alignment. Generally, both slit
widths will be set as 2.5nm.

15. Using the reflect mirror (PN FS2-ACCE-02), you can find the xenon lamp beam position
in the entrance slit of the emission monochromator.

16. Install the emission monochromator in the proper position, monitoring xenon lamp
beam position in the entrance slit of the emission monochromator. After the beam
alignment is completed, fasten the emission monochromator carefully for the beam
alignment not to be distorted. Fasten the socket head screws (total 4ea) crossed by the
slanting lines to prevent emission monochromator alignment from distorting.

The wavelength calibration of monchromators should be checked periodically. Especially
on monochromators where the ‘calibration’ is determined electronically rather than by
direct mechanical coupling. For calibration we use a low pressure Hg(mercury)-Ar (argon)
lamp with special accessories. This lamp provides a number of sharp lines which can be
used for calibration, and then measured Hg(mercury)-Ar (argon) wavelengths are
compared with the known values. A small slit width is used to increase precision of the
wavelength determination and to decrease the light intensity, which is important to
attenuate the light so that the photomultiplier tube and/or amplifiers are not damaged.

7. Find the zeroth order step value of the Emission monochromator to search the
start step position of the Hg spectrum easily. Commonly the start step position of the
Hg spectrum is displayed after about 2000 step from the zeroth order step.

To know the zeroth order step of emission monochromator, first check the grating
position.
Monochromator has zeroth order when gratin position is parallel to case wall.
To rotate the grating, use the grating mode of Alignment software .

③ Grating will move corresponding to entered step value. And moved step value is

displayed on window.
④ When grating position is parallel to the case wall, record the displayed step value.

spectrum calibration procedures.
Change Air status filter to 530nm filter and then set PMT voltage value about 500V
(300V=>500V) the reason of the change from Air to 530nm filter is to get rid of the
Hg spectrum line interference.
The reason of increasing PMT voltage value is to increase Ar spectrum intensity since
the 530nm filter optical density decrease Ar spectrum intensity.

10. Check the combo box of view grating step and enter proper step value in the EM start
Step nearby the end of Hg spectrum step value and then click Start .

X-1-2. Excitation Monochromator calibration

After the calibration of the emission monochromator, the excitation monochromator can be
calibrated with a new standard. The slit width of both the emission monochromator and the
excitation and should be set the same small value, consistent with the available light
intensity a dilute suspension of glycogen is placed in the cell holder to scatter the exciting
light. The emission monochromator is set to a certain wavelength. If the excitation
monochromator is properly calibrated, then the maximum intensity of the scattered light is
seen when the indicated wavelength is identical.

XI-1-1. Sensitivity Test Method

1. Turn on the main power switch on the rear panel of the system.

2. Turn on the sub main power switch at the bottom right side of the front panel of the
system.

3. Push the Light Source button (Red) to turn on light firstly and push the Control Board
button (White) to turn on the main board.

caution
- It is important to keep up the order during turning powers on:
#1 Main P/S Æ #2 Light Source Æ #3 Control Board
- If the control board is powered on at first, it could cause damage to the
electric board by the high voltage/current when the light source is on later.
- To turn off the system, you need to follow the procedures in reverse order
against the start up. (#3 Control Board Æ #2 Light Source Æ #1 Main P/S)
- Before turning off the main power switch, you need to wait for over 10 minutes
at least to protect the lifetime of lamp after turned off the light source switch.

4. Check whether all three LED lights at the left side of the front panel of the system are lit
on. If it is ready, warm up the system for about 20-30 minutes.

The development of methods to obtain excitation and emission spectra, corrected for
wavelength –dependent effects, has been the subject of numerous investigations.
Spectrum correction is performed to make it possible to measure a true spectrum by
eliminating instrumental response needed to perform spectrum correction. ‘

XII-1-1. Generating emission correction factors

One way to generate emission correction factors for your instrument is to scan the
spectrum of a standard lamp. Because the actual irradiance values of the standard lamp
over wavelength are known, dividing the irradiance values by the lamp spectrum results in
a set of relative correction values.

1. Prepare a Standard Lamp set including an optical fiber and a reflect mirror adaptor.

2. Place the reflect mirror adaptor in the cell holder in the sample compartment.

3. Turn on the Standard lamp (push the Red color button HALOGEN) and warming it up
about 20 minutes.

1. Click Correct Excitation Spectra (Rhodamine B) and then click OK when the ‘Please

XIII-1-1. How to execute Diagnostics Software.

1. Turn on the main power switch on the rear panel of the system.

2. Turn on the sub main power switch at the bottom right side of the front panel of
the system.

3. Push the Light Source button (Red) to turn on light firstly and push the Control
Board button (White) to turn on the main board.

Caution!
- It is important to keep up the order during turning powers on:
#1 Main P/S Æ #2 Light Source Æ #3 Control Board
- If the control board is powered on at first , it could cause damage to the electric
board by the high voltage/current when the light source is on later.
- To turn off the system, you need to follow the procedures in reverse order against
the start up. (#3 Control Board Æ #2 Light Source Æ #1 Main P/S)
- Before turning off the main power switch, you need to wait for over 10 minutes at
least to protect the lifetime of lamp after turned off the light source switch.

4. Check whether all three LED lights at the left side of the front panel of the system
are lit on. If it is ready, warm up the system for about 20-30 minutes.

5. Run ‘Diagnostics’ in the Luminous Software folder on Desktop.

6. Click OK after ‘Reading serial number: OK’ window is displayed.

XIV. Firmware Upgrade
This document is made for helping firmware updating
conveniently.

Commonly the firmware updating of main-board is necessary when the


function of instrument and the function of accessories are added or changed.

The firmware updation of accessories ard added or changed


(e.g Fast Filter, multi cell, rapid mix and so on.)

XIV-1-1. Installation of the Firmware Update Utility for main B/D update

1. Unzip FW_Downloader.zip file.

2. Double click Setup.exe in the FW Downloader folder.

1. Turn off power of the instrument and disconnect the power cable.

2. Remove white screw caps (total 10 ea) and Phillips head screws, and then lift up the
upper case.

3. Separate the accessory connector socket module from the sample compartment, lifting
up the sample compartment.

4. Pull out the Flash Rom using IC extractor.
