I-1. Introduction
Lumina fluorescence spectrometer is designed to provide a complete analysis system
for fluorescence, phosphorescence and chemiluminescence measurements.
All instruments will be tested and proven to the performance before the shipment
Preparation Done
You are responsible for providing an acceptable operating environment. Attention paid
to the operating environment well ensures the continued high performance of your
Lumina fluorescence instrument. The instrument warranty will be made void if the
equipment is operated under sub-standard conditions.
The relative humidity of the operating environment should be lower than Maximum
75% with no condensation. Operating the Lumina fluorescence in very low humidity
may result in the accumulation and discharge of static electricity that shortens the
life of electronic components. Operating at high humidity will produce condensation
and result in short circuits. Thermo Scientific recommends that your laboratory be
equipped with a temperature / humidity monitor. This will ensure that your laboratory
is always in conformance with the temperature and humidity specifications.
Temperature
Vibration
Ensure that lab benches are free from vibration. If possible, any equipment
generating vibration during the operation should be installed on the floor rather than
alongside the system on the workbench.
Work bench
The workbench must be stable and durable enough to support the total weight of
equipment. The worktops should be large enough to permit a free circulation of air
around each unit in the system. Remember to provide space for the computer,
monitor and printer. To avoid damage through spillage of the samples being
analyzed, the worktops should be covered with a material which is corrosion
resistant and impervious to liquids.
Power supply
All power supplies must be single phase AC voltage, three wire system (active,
neutral, ground) and should be terminated at an appropriate power outlet and/or
regulations imposed by local authorities responsible for the supply of electrical energy
to the workplace.
A grounded AC power supply must be used. The Lumina accepts a dual voltage of
and printer.
Avoid using power supplies from a source that may be subject to electrical or RF
interference from other services (large electric motors, elevators, and welders for
example).
Insurance
Transit damage can be obvious or concealed and in either case will be admitted by the
carrier only if it is reported within the terms of the carrier’s agreement. For any claims
against damage in transit, the following general rules apply;
- Before accepting delivery, you must inspect the packages for signs of obvious
damage. The nature of any obvious damage must be noted on the carrier’s waybill,
which then must be countersigned by a representative of the carrier
- Within the time limit stated in the terms and conditions of carriage, a further inspection
must be made for concealed damage. If any damage is found at this stage, the carrier
must be modified in writing. You must retain all packing material for subsequent
inspection by a representative of the carrier.
- ‘SHOCK WATCH vibration indicator label that helps you to aware of the damage level
are attached on Packing Boxes. If the indicator color is changed as red color, it means
the packing box could be impacted/damaged by the physical shock externally.
If it is happened, contact Thermo Scientific or sales representative immediately to hear
the next procedures.

Caution
A. Xenon arc lamp has an explosion hazard. Make sure the system’s power is off
and all AC power is disconnected from the system. Read and follow the cautions
presented below.
B. Xenon arc lamp has an explosion hazard. Wear goggles and protective clothing
when opening the lamp housing and when handling the lamp.
C. The lamp will remain extremely hot for approximately one-half hour after it has
been turned off. Do not touch the lamp or the metal unit until the lamp has cooled.
D. Never look directly at the xenon arc or its reflection. Intense radiation can
permanently damage eyes.
E. Do not touch the focusing lens, back scatter mirror, or the surface of the lamp.
Fingerprints will be burned onto these surfaces when the lamp is ignited.
1) Start
2) Turn off the power of the instrument.
3) Remove white screw caps (total 10) located around on the upper case.
4) Remove Phillips Head screws using a Phillips screwdriver.
5) Remove Phillips head screws (4 ea) located on the Sample Cell Assay part using a
Phillips screwdriver.
6) Separate the Sample Cell Assay part and lift the upper case.
7) Get rid of socket screws (4 ea) using a hexa socket screwdriver.
8) Separate the lamp connector from the lamp power module and push the hap and pull
out the connector.
9) Remove the lamp mounting module from the lamp housing.
10) Remove the screws (2 ea) located on the edges of the lamp.
11) Unfasten the socket screws (2ea) located on the (+) side of the Teflon bracket.
12) Unpack a new Xenon lamp.
13) Install the lamp in the correct polarity.
14) Fasten the socket screw (2ea) located in (+) direction Teflon bracket. Put the changed
lamp module back in the lamp housing.
15) Finally, connect the lamp cable to the power supply cable.

1. Check the main power off (If the system and lamp are on, it requires enough cooling
time of lamp over 10 minutes after turning off the control board and lamp. Then, turn off
the sub main power switch at the bottom right side of the front panel and the main
power on the rear panel)
2. Remove white screw caps (total 10 ea) located around on the upper case.
Caution:
Install the lamp in the correct polarity. Otherwise it could cause burst, overheating of the
lamp house or short lamp life. (Upper side is plus polarity)
13. Do not use any tools to tighten nuts when fixing the lamp, use hands only. Otherwise it
could cause breakage.
15. Fasten the socket screw (2ea) located in (+) direction Teflon bracket. Put the changed
lamp module back in the lamp housing.
16. Finally, connect the lamp cable to the power supply cable.

2) Turn off the power of the instrument.
3) Loosen the fuse holder on the rear side of the instrument.
4) Pull out the fuse (AC 250V, T2AL) and exchange it with a new one.
5) Put the fuse holder back and tighten.
6) End

2) Turn off the power of the instrument.
3) Pull out the fuse box on the rear side of the instrument.
4) Replace the fuse (AC 250V, T5AL) with a new one.
5) Put the fuse box back as it was.

1. Check the main power off (If the system and lamp are on, it requires enough cooling
time of lamp over 10 minutes after turning off the control board and lamp. Then, turn off
the sub main power switch at the bottom right side of the front panel and the main
power on the rear panel)
2. Loosen the fuse holder by turning it clockwise on the rear side of the instrument, using
(-) driver
3. Pull out the fuse (AC 250V, T2AL) and exchange it with a new one.
Caution: Should use the fuse of rated capacity (AC 250V, T2AL).

2. Pull out the fuse box on the rear side of the instrument as below.
Caution: Should use the fuse of rated capacity (AC 250V, T5AL).

2) Turn off the power of the instrument.
3) Check the Xenon lamp power supply’s location before starting replacement work.
4) Disconnect xenon lamp power cable, AC power connector and On/Off signal
connector.
5) Unfasten the socket screws (4ea) located on the base plate of the Xenon lamp
power supply.
6) Replace the Xenon lamp power supply with a new one and re-assemble in reverse
order.

1) Start
2) Turn off the power of the instrument.
3) Separate cables connected from the SMPS.Disconnect xenon lamp power cable,
AC power connector and On/Off signal connector.
4) Unfasten the socket screws (4ea) located on the base plate.Replace the Xenon
lamp power supply with a new one and re-assemble in reverse order.
5) After replace the SMPS, reconnect the cables to the new SMPS.

2) Turn off the power of the instrument.
3) Separate all connectors from the main controller Board.
4) Remove the Phillips screws (8ea) located on the main controller board bracket (the
red-colored marking point) and separate the main controller board from the
bracket.
5) Remove the Phillips screws (2ea) located on the step motor board from the main
controller board bracket (the red-colored marking point) and separate the step
motor board from the main controller board.
6) Remove the Phillips screws (2ea) located on the A/D board from the main
controller board (red-colored marking point) and separate the A/D board form the
main controller board.
7) Replace the main controller board and reassemble all the separated items (Step
motor board, A/D board, Cable connectors) in reverse order.
8) End
3. Remove the Phillips screws (8 ea) located on the main controller board bracket (the
red-colored marking point) and separate the main controller board from the bracket.
5. Remove the Phillips screws (2 ea) located on the A/D board from the main controller
board (red-colored marking point) and separate the A/D board from the main controller
board.
6. Replace the main controller board and reassemble all separated items (Step motor
board, A/D board, Cable connectors) in reverse order.
1) Start
2) Turn off the power of the instrument and check the location of the PD (Photo-
Diode) sensor board.
3) Separate all connectors, release the snap keys and lift the optical housing
connected with the PD sensor board.
4) Remove the Phillips screws (4ea) _ (red-colored marking point) located on the PD
sensor board and separate the board from optical housing.
5) Replace the PD Sensor Board with a new one and reassemble all the connectors
in reverse order.
6) End
1. Turn off the power of the instrument and check the location of the PD (Photo-Diode)
sensor board.
2. Separate all connectors, release the snap keys and lift the optical housing connected
with the PD sensor board.
3. Remove the Phillips screws (4 ea) (red-colored marking points) located on the PD
sensor board and separate the board from the optical housing.
1) Start
2) Turn off the power of the instrument and check the location of the Xenon
Lamp On/Off Check Board and lift the optical housing after releasing the snap keys.
3) Remove the socket screws (6ea) located on the optical base plate (red color
marking point) and lift the optical base plate.
4) Remove the Phillips screws (2ea) located on the optical housing which is adapted
to the On/Off Check Sensor Board, and lift the optical housing.
5) Remove the Phillips screws (4ea) located on the Xenon On/Off Check Sensor
Board (red color marking point) and Separate the Xenon On/Off Check Sensor
Board from the optical housing.
6) Reinstall a new Xenon On/Off Check Sensor Board in the optical housing and
connect all the connectors in reverse order.
7) End
1. Turn off the power of the instrument and check the location of the Xenon Lamp On/Off
Check Board and lift the optical housing after releasing the snap keys.
2. Remove the socket screws (6 ea) located on the optical base plate (red color marking
point) and lift the optical base plate.
5. Reinstall a new Xenon On/Off Check Sensor Board in the optical housing and connect
all the connectors in reverse order.
1) Start
2) Turn off the power of the instrument and check the location of the LED Display
Board.
3) Separate the cable connector and unfasten the nut screws (4ea), the bolt screws
(4ea) on the LED display Board.
4) Reinstall a new LED Display Board in the Front panel and connect all the
connectors in reverse order.
5) End
1. Turn off the power of the instrument and check the location of the LED Display Board.
2. Separate the cable connector and unfasten the nut screws (4ea) located on the LED
Display Board.
3. Reinstall a new LED Display Board in the Front panel and connect all connectors in
reverse order.
1) Start
2) Turn off the power of the instrument and check the location of the step motor board
for the slit and the filter module, and separate all connectors from the main
controller board.
3) Remove the Phillips screws located on the main controller board (red color
marking point) and separate the step motor board for the slit/filter module and the
step motor board for the grating module from the Main controller board.
4) Reinstall the new step motor boards and connect all the connectors in reverse
order.
5) End
1. Turn off the power of the instrument and check the location of the step motor board for
the slit and the filter module, and separate all connectors from the main controller
board.
2. Remove the Phillips screws located on the main controller board (red color marking
point) and separate the step motor board for the slit / filter module and the step motor
board for the grating module from the Main controller board.
3. Reinstall the new step motor board and connect all connectors in reverse order.
1) Start
2) Turn off the power of the instrument and check the location of the A/D board and
separate all the connectors from the main controller board.
3) Remove the Phillips screws located on the main controller board (red color
marking point) and separate the A/D Board from the main controller board.
4) Reinstall the new A/D boards with the main controller board and connect all the
connectors in reverse order.
5) End
1. Turn off the power of the instrument and check the location of the A/D board and
separate all connectors from the main controller board.
2. Remove the Phillips screws located on the main controller board (red color marking
point) and separate the A/D Board from the main controller board.
3. Reinstall a new A/D Board with the main controller board and connect all the
connectors in reverse order.
1) Start
2) Turn off the power of the instrument and check the location of the PMT sensor
board and separate all the connectors from the main controller board.
3) Remove the Phillips screws located on the main controller board for the PMT
Sensor board (red color marking point) and separate the PMT Sensor.
4) Reinstall a new PMT Sensor board in the main controller board and connect all the
connectors in reverse order.
5) End
1. Turn off the power of the instrument and check the location of the PMT sensor board
and separate all the connectors from the main controller board.
2. Remove the Phillips screws located on the Main controller for the PMT Sensor board
(red color marking point) and separate the PMT Sensor.
3. Reinstall a new PMT Sensor board in the main controller board and connect all the
connectors in reverse order.
1) Start
2) Turn off the power of the instrument and check the location of the Accessories
board and separate all the connectors from the main controller board.
3) Separate the sample compartment and check the position of accessories
connector panel.
4) Separate all the connectors from the main controller board and Accessories
connector panel.
5) Remove the screws located on the accessories board
6) Reinstall a new accessories board and connect all the connectors in reverse order
7) End
1. Turn off the power of the instrument and check the location of the Accessories board
and separate all the connectors from the main controller board.
2. Separate the sample compartment and check the position of accessories connector
panel.
3. Separate all the connectors from the main controller board and Accessories connector
panel.
5. Reinstall a new accessories board and connect all the connectors in reverse order.
Caution
A. Handle tubes with extreme care.
PMT has evacuated glass envelopes, allowing the glass to be scratched or subjected to
shock can cause cracks. Take extreme care during handling, particularly for tubes with
graded sealing on synthetic silica bulbs.
1) Start
2) Turn off the power of the instrument and check the location of the PMT Sensor
Board.
3) Separate all the connectors (red color marking point) from the main controller board.
4) Remove the Phillips screws (4ea) located on the PMT bracket (red color marking point).
5) Draw out the PMT carefully.
6) Reinstall a new PMT with the PMT sensor bracket and connect all the connectors in
reverse order.
7) End
1. Turn off the power of the instrument and check the location of the PMT Sensor board.
2. Separate all the connectors (red color marking point) from the main controller board.
Caution:
We recommend this method is performed in dark room for protecting PMT.
Caution
Check out the direction of the PMT socket assembly pin array before the reinstallation of
the PMT sensor in the PMT socket assembly.
1) Start
2) Turn off the power of the instrument.
3) Disassemble each housing part and connected cables. And Disconnect the Fan
connectors (2ea) and disassemble the rear panel to pull the excitation monochromator
out from the base plate.
4) Exchange the defected monochromator with a new monochromator.
And reassemble the cables and the housing part.
5) Monochromator Alignment.
6) Slit calibration, Filter calibration, and monochromator wavelength calibration.
7) End

2. Remove white screw (total 10) located around on the upper case.
7. Disconnect the worm drive step motor cable, the filter step motor cable, and the slit
step motor cable. Remember each pin position and label number for re-connection.
9. Pull monochromator out as shown in the right-picture below to release its connector
conveniently.
10. After the removal of the defected monochromator is completed, insert a new
monochromator to the base plate and place it in the suitable screw position.
Be careful to connect them to each the right position. If the cables are not connected
in the proper position, the components would not be operated properly.
13. Run the ‘Alignment.exe’ for alignment procedure located in Lumious Software folder
[Power supply]
The power supply of the xenon lamp is extremely dangerous, it generates the high
voltages so take a great care to the electric shock.
[Xenon lamp]
Xenon lamps emit ultraviolet radiation which is harmful to the eyes and the skin so
Always wear a light-shielding protector while the lamp is on.
[Grating]
1) Never touch grating surface. Crusts do not affect on optical characteristics in most
cases but fingerprints cause surface deterioration.
2) Do not wipe the sides of grating by solvent and /or clip the edges of grating plane.
When liquids and/or oil ingredients are adhered in the area of the edges of grating
[Air Fan]
Disconnect the cable of air blow fan for protection of grating during the service.
Oil of DC motor could attack the surface of grating.
16. 1) Place the upper optic module and screw socket head screw. (Refer to below
figure.)
2) Open the shutter.
3) Put the Teflon Block in the center of cell compartment. (Refer to below
figure.)

18. Reassemble the upper optical protect housing, the xenon lamp protect housing, the
side panel of monochromator, the upper case and the sample cell assay.
1) Start
2) Turn off the power of the instrument.
3) Disassemble each housing part and connected cables.
4) Exchange the defected monochromator with a new monochromator.
And reassemble the cables and the housing part.
5) Monochromator Alignment.
6) Slit calibration, Filter calibration, and monochromator wavelength calibration.

2. Remove white screw (total 10) located around on the upper case.
7. Disconnect the Fan connectors (2ea) and disassemble the rear panel to pull the
emission monochromator out from the base plate.
9. Pull out the PMTbulb from the PMT housing and reassemble it to a new PMT housing
of emission monochromator carefully.
Caution
We recommend this method is performed in dark room for protecting PMT.

the worm drive step motor, the filter step motor and the slit step. All the cables should
be connected in their proper positions.
12. Run the ‘Alignment.exe’ for alignment procedure located in Luminous Software folder
13. Turn on the xenon lamp, and Calibrate Slits and Filters using Alignment software.
After finishing calibration of slits and filters, set the filter as ‘air blank’ and the slit 2.5nm
and move the Excitation monochromator grating to zeroth order for beam alignment.
14. Install Teflon block in the cell holder. The Teflon block (PN FS2-ACCE-08) reflects the
xenon lamp beam from the excitation lens holder to the emission lens holder and that
will be helpful to make an emission monochromator alignment. Generally, both slit
widths will be set as 2.5nm.
16. Install the emission monochromator in the proper position, monitoring xenon lamp
beam position in the entrance slit of the emission monochromator. After the beam
alignment is completed, fasten the emission monochromator carefully for the beam
alignment not to be distorted. Fasten the socket head screws (total 4ea) crossed by the
slanting lines to prevent emission monochromator alignment from distorting.
18. Find each proper slit position of the emission monochromator, using the reflect mirror,
xenon lamp beam and the ‘Alignment’ software program.
1) Start
2) Turn off the power of the instrument. And separate sample compartment.
3) Disassemble each housing part and cables.
4) Exchange the defected shutter module with a new shutter module.
And reassemble sample cell assay part and the upper case in reverse order.

3) Disassemble each housing part and cables.
4) Exchange the defected shutter module with a new shutter module.
And reassemble sample cell assay part and the upper case in reverse order.

the shutter module cable which is connected to the main board.

3) Unfasten the socket screw (2ea)
4) Disconnect the cables.

7. Disconnect the cables that correspond to power switch module from Terminal block,
Noise filter, Xenon lamp power, SMPS power supply.
8. Exchange the defected power switch module with a new power switch module

3) Install Hg-Ar lamp to the instrument.
4) Make Emission Monochromator Calibration
5) Remove the Hg-Ar lamp from the instrument.
6) Place the Teflon Block into cell holder.
7) Make Excitation Monochromator Calibration.

1. Prepare a low pressure Hg(mercury)-Ar (argon) lamp, a fiber optic and a fiber adaptor.
This Lamp lifetime is only about 50 hours. Therefore, you should always turn off
the power of the lamp after the calibration procedure is finished.

To know the zeroth order step of emission monochromator, first check the grating
position.
Monochromator has zeroth order when gratin position is parallel to case wall.
To rotate the grating, use the grating mode of Alignment software .
② Enter proper step value you want and click the arrow mark.
③ Grating will move corresponding to entered step value. And moved step value is
displayed on window.
④ When grating position is parallel to the case wall, record the displayed step value.
Tip: You can find the approximate zeroth order step referring to the
‘Alignment.ini’ file located in the Luminous Folder.
10. Check the combo box of view grating step and enter proper step value in the EM start
Step nearby the end of Hg spectrum step value and then click Start .
13. After all step value peaks of Hg & Ar spectrum were recorded in the table,
Click calibration, and then the ‘Curve fitting is OK’ window is displayed.
After the calibration of the emission monochromator, the excitation monochromator can be
calibrated with a new standard. The slit width of both the emission monochromator and the
excitation and should be set the same small value, consistent with the available light
intensity a dilute suspension of glycogen is placed in the cell holder to scatter the exciting
light. The emission monochromator is set to a certain wavelength. If the excitation
monochromator is properly calibrated, then the maximum intensity of the scattered light is
seen when the indicated wavelength is identical.
2. Place the Teflon block into the cell holder in the sample compartment.
4. Click New folder and save it with proper file name when save window is displayed

click OK and Save.
1. Turn on the main power switch on the rear panel of the system.
2. Turn on the sub main power switch at the bottom right side of the front panel of the
system.
3. Push the Light Source button (Red) to turn on light firstly and push the Control Board
button (White) to turn on the main board.
caution
- It is important to keep up the order during turning powers on:
#1 Main P/S Æ #2 Light Source Æ #3 Control Board
- If the control board is powered on at first, it could cause damage to the
electric board by the high voltage/current when the light source is on later.
- To turn off the system, you need to follow the procedures in reverse order
against the start up. (#3 Control Board Æ #2 Light Source Æ #1 Main P/S)
- Before turning off the main power switch, you need to wait for over 10 minutes

One way to generate emission correction factors for your instrument is to scan the
spectrum of a standard lamp. Because the actual irradiance values of the standard lamp
over wavelength are known, dividing the irradiance values by the lamp spectrum results in
a set of relative correction values.
1. Prepare a Standard Lamp set including an optical fiber and a reflect mirror adaptor.
2. Place the reflect mirror adaptor in the cell holder in the sample compartment.
