The structural and functional unit of life is cell, Robert Hooke, 1665 A.D. observed the cellfor the first time.
Purpose : Tostudy the plant cell by preparing temporary stained slide of onion bulb peel.
Requirement: Onion, slide, cover slip, needle, razor, iodine solution, microscope, glycerine etc.
Procedure
: After peeling the onion from its dry leaves, take out one fleshy leafy leaf and from its inner
side remove its thin membranous peel and place in water filled watch glass.
Makesmallpieces of this peel and place on the slide and add adrop of iodine solution. Now
cover it with cover slip and place under microscope for further investigation.
Observation : Rectangular polygonal cells which are closely packed are seen. Cells are surrounded by
distinct cell wallwith thin cell membrane on inner side. Dark coloured nucleus may also be
spotted with large vacuoles.

Unit 5 Animal Life (Microscopic Study)
Purpose :To study the temporary stained slide of cheek epithelial cells of man.
Requirement : Toothpick, slide, methylene blue, cover slip, watch glass, etc.
Procedure : Wash your mouth properly and scrap the internal lining of cheek with acean tooth pick
gently. keep the scrapping on aclean slide in adrop of saline solution and add adrop of
methylene blue.
After 2-3 minutes remove the skin and add a drop of glycerine on the scrapping and cover
it with cover slip.
Observation : Large number offlat, irregular, polygonal, leafy cells are seen.
Cells have distinct nucleus in the center.
Cytoplasm is grannular and rough.
