Afterwards the class will be grouped into 3 to
practice making wet mounts. The materials like a
printed paper, scissors, dropper, glass slide, cover
slip and compound microscope will be given. The
student will be given learning notes also as their
guide.

Part I: Making Wet Mount


1. Cut out a small letter “e” from a printed paper. Using
forceps or tweezers place it in at the center of a glass slide
in an upright position.
2. Add a drop of tap water over the specimen. It will act as a
mounting medium and make clear the image of the
specimen. Position the cover slip at 45° with one side
touching one edge of the water on the slide.
3. Slowly lower the other edge of the cover slip until it rests
on the water and the printed letter. Bubbles are perfect
circles you see on your preparation. Remove or minimize
trapped bubbles by gently tapping the cover slip with the
eraser-end of a pencil. Make the bubble move towards the
edge of the cover slip.

Part 2: Observing Specimen


1. Put the slide on the stage. Make sure that the letter “e” is
at the center of the hole in the stage and under the LPO.
Hold it firmly with the stage clips.
2. Watching from the side, carefully lower the body tube
until the end of the LPO almost touches the cover slip.
3. Look through the eyepiece. Slowly turn the coarse
adjustment upwards to raise the objective until the letter “e”
appears. Continue until you see the letter clearly. This
would indicate that you have focused it already.

multiplying the number found in the eyepiece with the
number found on the objective being used. So, if a
specimen is viewed using a 10x objective and a 10x
eyepiece it will be magnified 100 times.

SCAN IT!, ILLUSTRATE IT! and ORGANIZE IT!

1. SCAN IT! (Tier 3)


Students will be asked to focus an onion cell under
the scanner and LPO and present it to the class
using the camera of the phone casted to the TV
monitor for others to see.

2. ILLUSTRATE IT! (Tier 2)


Students will be sketching the image of an onion
cell seen with and without stain under the LPO.
(MAPEH-across curriculum)
3. ORGANIZE IT! (Tier 1)
Students will be asked to demonstrate how to focus
an onion cell from LPO to HPO while identifying
the most significant part of the cell seen under the
microscope.

Each group will be given an onion, blade, microscope, glass


slide, iodine solution, tissue and Learning module as their
guide.
*The teacher will remind the student to be careful in using
the blade so as not to cut their skin or to avoid injury.

Part 3: How to prepare a stain specimen


1. Cut the onion into quarters.
2. Remove one fleshy scale and detach carefully the
transparent skin from the inner surface of the onion
scale using the tweezer.
3. Place the thin transparent onion skin on the glass
slide with the inner surface (non-waxy slide) facing
up. Make sure that it is not folded or wrinkled.
4. Add a droplet of distilled water on the onion skin
and cover it with the slip.
5. Observe the onion cell under the microscope
shifting from the scanner to HPO.
6. Remove the slide from the stage. Using a dropper
add one or two drops of iodine solution along the
edge of the cover slip. Remove the extra liquid
from the edge of the cover slip using a tissue paper.
7. Place the slide on the stage and observe the
specimen from the scanner to LPO and HPO.
8. Record your observation of the onion slide with and
without a stain.
9. Clean the microscope after using.
10. Prepare to present the task given.
