There are three main types of gene transfer in bacteria: 1) Conjugation - transfer of genetic material between bacterial cells through cell-to-cell contact via pilus structures. 2) Transduction - transfer of genes from one bacterium to another via bacteriophages during viral replication. 3) Transformation - uptake and integration of naked DNA from the environment into competent bacterial cells.

Gene transfer in bacteria

• There are three types of gene transfer in bacteria by which donor DNA

is transferred into a recipient cell.

1) CONJUGATION
• Bacterial conjugation is the transfer of genetic material between bacterial cells

through pili.

Steps in conjugation:
1. Donor cell produces pilus.
2. Pilus attaches to recipient cell and brings the two cells together.
3. The double stranded plasmid is converted into single strand which is then
transferred to the recipient cell.
4. Both cells synthesize a complementary strand to produce a double stranded
circular plasmid and also produce pili; both cells are now donor for the F factor.

• Donor cells are called F+ 

• Recipient cells are called F- 

• When F- cells receive plasmid they become F+ and then can

donate a copy of plasmid.

2) TRANSDUCTION
• Transduction is the process by which foreign DNA is introduced

into a bacterial cell by a virus.


Two cycles of virus during transduction

1) The lytic cycle: It involves the reproduction of viruses using a

host cell to manufacture more viruses; the viruses then burst out

of the cell.

2) The lysogenic cycle: It involves the incorporation of the viral genome into the

host cell genome and it replicates along with it harmlessly. Integrated Viral DNA

is called prophage.

The lysogenic cycle does not result in immediate lysing of the host cell. When

nutrients in bacterial cells are finished, viral DNA (prophage) can become active,

reproduce and finally cause lysis of bacterial cell.


3) TRANSFORMATION
• A process by which some bacteria take up foreign genetic

material (naked DNA) from the environment.

• Bacteria that can intake naked DNA from environment are

called competent cells.

• It was first reported in Streptococcus pneumoniae by

Griffith in 1928.
